My lab focuses on inner ear regeneration through stem cell-based approaches. In vitro, we developed a co-culture system to investigate whether mouse ESCs-derived neurons synapse with mouse auditory neurons. The newly formed synapse-like structures express both pre- and post-synaptic proteins. Functional assays, including synaptic vesicle recycling, paired recordings, and blocker electrophysiology, suggest the presence of active synaptic vesicles, transsynaptic activity, and the formation of functional glutamatergic synapses. In vivo, DNA demethylation may contribute to the regeneration of auditory hair cells following chemically-induced deafness. Our data highlight the potential of stem cells in repairing the damaged auditory system.
