Focal adhesion kinase (FAK) is recognized as a key downstream mediator of integrin signaling, promoting cell migration, proliferation, and survival. For the first time, we have discovered that FAK is present in the nucleus and catalytically inactive in healthy vascular cells, including endothelial cells, smooth muscle cells, and cardiomyocytes in vivo. Our work further suggests that nuclear FAK plays a role in epigenetic and transcriptional regulation. Expanding beyond vascular disease, we are also exploring the emerging roles of nuclear FAK in cancer.
