Arterial stiffening is a hallmark of aging and a key determinant of cardiovascular risk. One of the main elements of arterial stiffening is remodeling of the vascular ECM. Lysyl oxidases, the family of amine oxidases, play an important role in establishing and maintaining the vascular matrix. However, dysfunctional activation of these enzymes leads to derangements in the structural and functional parameters in the vasculature, resulting in maladaptive stiffening. The role of these enzymes in vascular health and disease will be discussed from the molecular level to physiological impact.
