Beige adipocytes in inguinal white adipose tissue (iWAT) are essential for adaptive thermogenesis. We identify estrogen receptor α (ERα) signaling in tissue-resident PDGFRα⁺ cells as a key regulator of beige adipogenesis. Loss of ERα impairs cold-induced beige fat formation, increases fibrosis, and reduces thermogenic capacity through enhanced TGFβ signaling. Pharmacological inhibition of TGFβ with CJJ300 restores beige adipogenesis and thermogenic function in ERα-deficient and aged mice, highlighting ERα–TGFβ crosstalk as a critical pathway regulating beige fat plasticity.
