Primary human fibroblasts can use at least two distinct mechanisms or modes of migration through three-dimensional (3D) matrices. We now find that this switch in migration mode is accompanied by significant changes in the distribution and dynamics of mitochondria in the migrating fibroblasts. We propose that perinuclear, stationary mitochondria generate the additional ATP required for fibroblasts to power the movement of the nucleus within cells migrating through crosslinked extracellular matrices.
