RNA modifications emerge as a critical mechanism to govern cell proliferation, differentiation, and function. METTL14 is an essential component of the N⁶-methyladenosine (m⁶A) transferase that deposits m⁶A methylation on RNA. YTHDC1 specifically binds m⁶A -methylated RNA to regulate RNA fate. We generated and characterized genetically modified mice with deletion of Mettl14 or Ythdc1 in adipocytes, hepatocytes, or neurons. Our results demonstrate that the METTL14/ m⁶A /YTHDC1 epitranscriptomic pathway shapes the activities of these cells and is indispensable for the maintenance of energy balance and metabolic homeostasis.
