Abstract: HIV-1 engages innate immune sensing during the early stages of infection, with PQBP1 targeting the viral capsid and facilitating cGAS recognition of newly synthesized viral DNA in myeloid cells. We investigate how this sensing axis operates in monocyte-derived dendritic cells and microglia and how it is modulated by antiretroviral therapies. Our findings link the recognition of incoming HIV-1 particles to sustained cGAS–STING activation, inflammatory signaling, and disrupted neural homeostasis in microglia-containing brain organoids and humanized mouse xenograft models.
